AI methods can predict protein structures, propose backbones, and help select sequences.

These are research tools: predicted structure or designed sequence alone does not establish biological function, therapeutic effect, safety, or clinical readiness.
AlphaFold predicts protein structures from sequence, while methods such as RFdiffusion propose protein backbones and ProteinMPNN predicts sequences compatible with a target backbone. The cited RFdiffusion and ProteinMPNN studies report particular computational and laboratory evaluations; their results do not establish that any generated candidate is functional or suitable as a medicine. Antibody CDR loops contribute to antigen binding, but successful binding, specificity, developability, and safety require experimental evaluation. AlphaFold 3’s developer description includes modeling interactions involving proteins, DNA, RNA, small molecules, ions, and modified residues; predictions remain research hypotheses. For a therapeutic biologic intended for human use and marketing in the United States, FDA describes preclinical and clinical development and applicable marketing review; recombinant therapeutic proteins may require a biologics license application. This pathway does not apply to every research protein or to non-therapeutic structures. AlphaFold-Multimer is a separate model variant developed for predicting structures of multiple interacting protein chains; those predictions are hypotheses that require experimental assessment. The associated primary paper reports improved interface prediction in its evaluated settings.
AlphaFold’s 2020 publication describes protein-structure prediction from amino-acid sequence.
RFdiffusion’s paper evaluates computationally designed protein structures and selected experimental validations.
ProteinMPNN predicts sequences for target backbone structures in reported design experiments.
Illustrative: researchers may test candidate antibody loops experimentally; no design method guarantees binding or therapeutic performance.
A useful research direction is to measure how computational design success transfers to expression, stability, binding, and functional assays. Therapeutic development requires product-specific evidence and the applicable regulatory pathway; do not infer clinical benefit from a computational design result.

ProteinMPNN is an inverse-folding model: it predicts candidate sequences for a specified backbone. A prediction does not guarantee that a sequence will fold or function experimentally.
